FORMULASI DAN OPTIMASI KADAR HYDROXYPROPYL METHYLCELLULOSE K100M DAN POLYVINYL ACETATE DALAM SEDIAAN PATCH TRANSDERMAL PROPRANOLOL HCl

چکیده

Propranolol hidroklorida merupakan penghambat reseptor beta-1 yang digunakan untukterapi angina, aritmia jantung, dan hipertensi. Penggunaannya secara oral memiliki beberapamasalah seperti mengalami first pass effect, indeks terapi sempit, rasa pahit.Kekurangan tersebut bisa diatasi dengan memformulasikannya sebagai sediaan transdermalberupa patch. Komponen dalam patch transdermal adalah kombinasipolimer hidrofilik yaitu HPMC K100M PVA. Optimasi dilakukan SimplexLattice Design agar diperoleh formula optimal menghasilkan transdermalpropranolol HCl karakteristik fisik daya penetrasi terbaik. Penelitian inibertujuan mengetahui pengaruh konsentrasi PVA pada karakteristiksediaan propranolol serta mencari konsentrasioptimal. perbandingan ditentukanmenggunakan Expert 11.0.1 metode Simplex Lattice untuk 8 formulauji. Masing-masing diuji meliputi pH, keseragaman bobot,ketahanan lipat, ketebalan uji sehingga optimalkemudian validasi SPSS melalui one sample t-test. Hasil penelitianmenunjukkan ada kombinasi dandaya HCl. Formula optimum berdasarkanDesign dayapenetrasi paling baik yakni 6,13% 2,37%. Kata kunci : patch, hidroklorida, K100M, Design,
